Effect of Yiqi Bushen Recipe on expression of notch1 and jagged1 in tumor cells of mouse gastric cancer model

Abstract
OBJECTIVE: To explore the molecular mechanism of multiple targets of Yiqi Bushen Recipe (YBR) in resisting gastric cancer metastasis. STUDY DESIGN: A cancer stem cell (CSC)–enriched, tumor-bearing nude mouse model was established by using luciferase-labeled human gastric cancer cell line SGC-7901-luc. The experimental mice included 5 groups: negative control (group 1), model control (group 2), 5-fluorouracil injection (5-FU group 3), YBR (group 4), and YBR+5-FU (group 5). The drug intervention time was 6 weeks (groups 3, 4, and 5). The protein expressions of Notch homolog 1 (Notch1) and Jagged1 (Jag1) were detected by western blot. The mRNA expressions of Notch1 and Jag1 were detected by RT-PCR. RESULTS: The Notch1 and Jag1 protein concentrations in the groups 4 and 5 were lower than those in group 2 (p<0.05). The level of Notch1 and Jag1 mRNA was lowest in group 5, followed by group 4, and their concentrations were significantly different from group 2 and group 3 (p<0.05). CONCLUSION: The mechanism of YBR against gastric cancer (GC) metastasis may be related to the down-regulation of the protein and mRNA expression of Notch1 and Jag1. It is beneficial for patients with GC to take YBR. © Science Printers and Publishers, Inc.
